马鞭草苷通过Wnt/β-catenin信号通路干预哮喘大鼠气道炎症及气道重塑的研究 被引量:16

摘要 目的探讨马鞭草苷对哮喘大鼠气道炎症、气道重塑及Wnt/β-catenin信号通路的影响。方法将50只SD大鼠随机分为正常(Con)组、模型(Mod)组、马鞭草苷低(VL)、中(VM)、高(VH)剂量组,每组10只。除正常组外,其余大鼠采用卵清白蛋白(OVA)致敏建立哮喘模型,各组大鼠于致敏次日开始灌胃相应药物,末次激发2 h后进行支气管肺泡灌洗液(BALF)白细胞计数,酶联免疫吸附法(ELISA)检测BALF中IL-4、IL-5、TNF-α水平,观察大鼠肺组织病理形态改变,Western blot检测β-连环蛋白(β-catenin)、糖原合成酶激酶-3β(GSK-3β)的蛋白表达。结果与Con组比较,Mod组大鼠BALF中白细胞数、IL-4、IL-5、TNF-α水平增高,支气管壁厚度、平滑肌厚度增加,β-catenin表达升高,GSK-3β表达减少(P <0.05);与Mod组相比,VM、VH组大鼠BALF中白细胞数、IL-4、IL-5、TNF-α水平降低,支气管壁厚度、平滑肌厚度减小,β-catenin表达减少,GSK-3β表达增加,差异具有统计学意义(P <0.05)。结论马鞭草苷对哮喘大鼠的气道炎症和气道重塑具有抑制作用,其机制可能与调控Wnt/β-catenin信号通路有关。 Objective To investigate the effects of verbenalin on airway inflammation,airway remodeling and Wnt/β-catenin signal transduction pathway in asthmatic rats.Methods Fifty SD rats were randomly divided into normal control group(Con),model group(Mod),low(VL),medium(VM)and high dose group(CBH)of verbenalin.Animal models of asthmatic were induced by ovalbumin(OVA).Then different drugs were administrated via gavage after sensitization.After the last challenge,total cellular score and cellular classification in bronchoalveolar lavage fluid(BALF)were observed,the contents of IL-4,IL-5,TNF-αwere detected by ELISA.Pathological changes in the liver tissues were observed.The protein expressions ofβ-catenin and GSK-3βwere analysed by Western blot.Results Compared with the normal control group,the total cellular score,levels of IL-4,IL-5,TNF-α,bronchial wall thickness and smooth muscle thickness in the model group were significantly increased.The expression ofβ-catenin was reduced while the expression of GSK-3βwas increased(P<0.05).Compared with the model group,the total cellular score,levels of IL-4,IL-5,TNF-α,bronchial wall thickness,smooth muscle thickness and the expression of GSK-3βwere all reduced,and the expression ofβ-catenin was increased with significant differences(P<0.05).Conclusion Verbenalin can inhibit airway inflammation and airway remodeling probably by way of inhibiting the Wnt/β-catenin signal transduction pathway.
