摘要
目的本研究检测口腔扁平苔藓(OLP)及正常黏膜组织中MMP2和MMP9的表达情况,并在体外OLP细胞模型中进行激光照射下的MMP2和MMP9表达状况的检测,旨在探索其可能的作用机制,以期为激光治疗OLP提供临床及实验依据。方法 (1)应用免疫组化法和RT-PCR法测定20例OLP及20例正常口腔黏膜组织中MMP2、MMP9的表达水平;(2)激光处理上皮细胞Ha Cat cell,Western blot检测MMP2、MMP9蛋白的表达。结果 (1)MMP2在OLP中表达弱阳性,而在正常黏膜组织中几乎无免疫组化着色;MMP9在OLP中强烈表达于表皮角质形成细胞及真皮淋巴细胞胞浆,其表达显著高于正常黏膜组织(P<0.05);(2)激光呈时间和剂量依赖性下调上皮细胞Ha Cat cell MMP2、MMP9的表达。结论 (1)MMP2、MMP9表达在扁平苔藓患者中高于正常黏膜,提示MMP2、MMP9与OLP发病有关;(2)激光可下调体外OLP细胞模型中MMP2、MMP9的表达,提示激光可用于临床治疗OLP。
Objective To investigate the expression of MMP 2 and m mp9 in oral lichen planus(olp)and normal mucosa tissue,and to detect the expression of MMP 2 and m mp9 under laser irradiation in vitro olp cell model,in order to explore its possible mechanism of action and provide clinical and experimental basis for laser treatment of olp.Methods(1)The expression levels of MMP 2 and m mp9 in 20 olp and 20 normal oral mucosa tissues were determined by immunohistochemistry and rt-PCR.(2)The expression of MMP 2 and m mp9 protein was detected by laser treatment of ha cat cell and western blot.Results(1)MMP 2 was weakly positive in olp,but almost no immunohistochemical staining was found in normal mucosal tissues.M mp9 was strongly expressed in the cytoplasm of epidermal keratinocytes and dermal lymphocytes in olp,and its expression was significantly higher than that of normal mucosal tissues(P<0.05).(2)The expression of MMP 2 and m mp9 in epithelial cells ha cat cell was down-regulated by laser in a time-and dose-dependent manner.Conclusion(1)The expression of MMP 2 and m mp9 in lichen planus is higher than that in normal mucosa,suggesting that MMP 2 and m mp9 are related to olp.(2)laser can down-regulate the expression of MMP 2 and m mp9 in olp cell model in vitro,suggesting that laser can be used in clinical treatment of olp.
