摘要
为了解陕南地区白山羊泰勒虫的感染情况,采用形态学和分子生物学相结合的方法对121份来自陕西省陕南地区白山羊的血液样品进行检测。血液涂片染色后形态学检测结果表明,112份血液涂片的红细胞中存在大小约1μm,呈圆点形、卵圆形或短杆状多形性虫体。基于泰勒虫SSU rRNA基因位点对提取的血液样品DNA进行套式PCR扩增。结果表明,陕南地区白山羊的血液样品中泰勒虫阳性率为92.6%(112/121),且该结果与形态学检测结果相一致。通过比对发现,所获得的泰勒虫分离株与获取自GenBank中的吕氏泰勒虫相应基因序列序列同源性在99%以上。进一步构建遗传进化树发现,获得的泰勒虫分离株与吕氏泰勒虫(登录号:JN676988、AF081136)归于同一支,提示陕南地区白山羊感染的泰勒虫种为吕氏泰勒虫。
To systematically investigate the infection of Theileria spp.in Shaannan white goats,a total of 121 blood samples were collected from Shaanxi province and examined by using the morphological and molecular methods.Based on the microscopic examination,the polymorphic parasites were found in 112 blood samples with the round,oval and rod shapes and the size of approximate 1μm in the erythrocytes.Based on the nested PCR targeting SSU rRNA,a total of 112(92.6%,112/121)samples of Shaannan white goats were positive for Theileria infection,and the molecular results were in accordance with the morphological results.By sequence comparison,there was over 99%sequence identity between the sequences derived from the amplification products and the sequences of Theileria lunwenshuni obtained from GenBank.Further study based on the phylogenetic analysis found that all Theileria isolates and T.lunwenshuni(GenBank accession number:JN676988,AF081136)were grouped into a one clade,suggesting that Theileria isolates in this study was T.luwenshuni.
