荷斯坦牛BoLA-DRA基因外显子1生物信息学分析 被引量:2

摘要 本研究通过DNA混合池扩增测序方法对荷斯坦牛BoLA-DRA基因外显子1突变位点进行分析,并对其结构功能进行生物信息学预测。结果发现在荷斯坦牛BoLA-DRA基因外显子1在第1位和第2位之间有碱基T的插入,生物信息学预测结果表明DRA外显子1的mRNA二级结构突变前后自由能没有发生变化,蛋白质二级结构中β-折叠占比例最高,蛋白质功能预测结果显示该基因荷尔蒙和免疫应答的几率较高,表明DRA外显子1在荷斯坦牛性激素和免疫应答过程可能发挥重要作用,本研究结果为进一步研究BoLA-DRA基因抗病选育提供理论基础。 The mutation of exon 1 in Holstein BoLA-DRA gene were analyzed via PCR and DNA sequencing analysis,their structural function were predicted by bioinformatics in the study.The result found that there was an insertion of a baset T between the 1th and 2nd of the BoLA-DRA gene in Holstein cattle,and the bioinformatics prediction showed that the free energy of the mRNA secondary structure of the DRA exon 1 was unchanged before and after the mutation.The beta-folding was the highest in protein secondary structure.The results of protein function prediction showed that the hormones and immune response were higher,indicating that the DRA exon 1 might play an important role in the sex hormones and immune response process in Holstein,the results of research provides a theoretical basis for further study of BoLA-DRA gene resistance breeding.
