Pathogenic Detection and Phylogenetic Analysis of NDV in Four Kinds of Wild Birds of Guangxi During 2016—2017

摘要 为研究广西野生鸟类中的新城疫病毒(NDV)感染状况,从4种临床健康鸟类中,采集口咽/泄殖腔棉拭子650份,通过病毒分离、RT-PCR和F基因序列测定,进行NDV感染状况调查和分子特征分析。结果显示:共分离到10株NDV,分离率为1.54%;10株分离株F基因的氨基酸裂解位点基序均为^(112)R-R-Q-K-R-F^(117),符合NDV强毒株的典型特征;分离的10株毒株中,5株来源于斑鸠,3株来源于鸽子,2株来源于鹧鸪,8株为Ⅱ类NDV基因VI型,2株为Ⅻ型。结果表明,我国野鸟中流行的NDV以Ⅱ类基因Ⅵ型强毒株为主,但已出现了新基因型(基因Ⅻ型)。因此,应加大对野鸟NDV监测的力度,防止其将病毒传染给家禽,同时应开展当前疫苗对NDV Ⅻ型的免疫效果评估。 In order to study the infection status of Newcastle disease virus(NDV)of wild birds in Guangxi,650 oropharyngeal or cloacal swabs were collected from 4 kinds of clinically healthy birds.By virus-isolation,RT-PCR and F gene sequencing,investigation of NDV infection and molecular characterization were carried out.The results showed that 10 strains of NDV were isolated,and the isolation rate was 1.54%.The amino acid cleavage site motifs of the F genes in 10 NDV strains were all 112R-R-Q-K-R-F117,which was consistent with the typical characteristics of virulent NDV strain.Among the 10 isolated strains,5 strains were isolated from turtledoves,3 strains were isolated from pigeons,and 2 strains were isolated from francolinus pintadeanus.Besides,8 isolates belonged to genotype VI of class-ⅡNDV,and 2 isolates belonged to genotype XII.Results showed that the most prevalent strain of NDV in China was genotype VI,but a new genotype XII emerged.Hence,the surveillance for NDV in wild birds should be strengthened to avoid the domestic poulty being infected.Meanwhile,immune effect evaluation should be conducted towards the vaccine of class-ⅡNDV.
