摘要
目的:探讨不同剂量链脲佐菌素(STZ)腹腔注射建立昆明小鼠糖尿病模型的效果及稳定性。方法:选取雄性昆明小鼠32只,随机分为对照组和3个STZ组(A组、B组、C组),每组8只。小鼠适应性喂养一周后,连续5天腹腔注射不同剂量STZ(A组:40mg/kg·d;B组:50mg/kg·d;C组:60mg/kg·d)。造模1周后检测小鼠成模率及成模小鼠禁食2h、4h、6h、8h血糖。造模后每周检测对照组和成模小鼠血糖和体重。结果:C组连续5天腹腔注射能较好地诱导小鼠糖尿病的发生(成模率87.5%),且血糖水平稳定,死亡率较低。随着禁食时间的延长,小鼠血糖明显下降,禁食6h时,即有50%的小鼠血糖降至11.1mmol/L以下。结论:STZ 60mg/kg·d多次腹腔注射是诱导KM小鼠糖尿病的最佳剂量,且血糖水平稳定,死亡率低。
Objective:To investigate the effect and stability of STZ intraperitoneal injection to establish diabetes model with different doses.Methods:32 KM mice were randomly divided into normal control group and model groups(A,B,Cgroup),each group had 8 mice.Mice took back from animal house and maintain feed for a week,then mice of the model group treated with different doses STZ(A:40mg/kg·d,B:50mg/kg·d,C:60mg/kg·d)for 5 consecutive days.And the mice normal control were treated with citrate buffer.One week later,the model rate of mice was detected and the fasting 2h,4h,6h and 8h blood glucose were detected.Before treated with STZ,every mouse’s weight was measured.After treated with STZ,the blood glucose and weight of the normal control group and the model group were measured every week for 9 weeks.Results:The multiple intraperitoneal injection of STZ 60mg/kg·d can induce KM mice into diabetes(87.5%),and the blood glucose level was stable and the mortality was low.With the prolongation of the fasting time,the blood glucose of KM mice decreased significantly.When the fasting was 6h,50%mice’s blood glucose decreased to less than 11.1mmol/L.Conclusion:60mg/kg·d intraperitoneal injection of STZ is the best dose to induce diabetes in KM mice,and the blood glucose level is stable and the mortality rate is low.
