摘要
通过对牛支原体vspY蛋白基因的扩增、克隆及序列分析,研究vspY蛋白基因的保守性。结果:扩增vspY基因片段长度约为1 121 bp,vspY基因克隆测序显示基因片段确定为1 121 bp,与引物设计扩增长度相符;vspY基因序列分析显示,与牛支原体参考株基因同源性为98%,仅在533、728位点存在点突变;系统进化树显示,牛支原体贵州分离株与参考株处于不同系统进化分支,存在一定进化距离。结果表明:牛支原体vspY蛋白基因相对保守,不同地区分离株存在一定进化关系。
vspY gene was amplified,cloned and sequenced for analyzing the gene conservative of vspY.The results showed that the length of the amplified vspY gene fragment was 1 121 bp,same to the gene sequence length.The sequence analysis of vspY gene showed the genetic homology was 98%between bovine mycoplasma Guizhou strain and reference strains.There were two mutational sites at 533 and 728.Phylogenetic analysis showed bovine mycoplasma Guizhou strain was in different phylogenetic branches with reference strains and there were evolutionary distance between them.The results indicated vspY gene of bovine mycoplasma was conservative and had evolutionary distance between the strains isolated from different area.
