列当中黄药苷与牛血清白蛋白相互作用的荧光光谱研究

摘要 目的:研究列当中黄药苷(crenatoside)与牛血清白蛋白(bovine serum albumin,BSA)相互作用的荧光光谱,以期探讨二者的作用机制,为黄药苷的研究提供理论依据。方法:在模拟生理条件下,利用荧光猝灭、同步荧光技术,测定不同温度下随着黄药苷浓度的增大对BSA的淬灭过程。结果和结论:黄药苷对BSA有较强的猝灭作用,属于静态猝灭。常温下黄药苷与BSA的猝灭常数为5.14×1012L·mol-1;结合常数为2.95×105L·mol-1;结合位点数为1.17。根据热力学数推测了黄药苷与BSA的作用力类型;同步荧光的结果表明黄药苷对BSA的构像影响较小。 To research the fluorescence spectra of the interaction between crenatoside in Orobanche coerulescens steph and bovine serum albumin(BSA),in order to discuss the action mechanism of the crenatoside and BSA,and to provide theoretical basis for the study of crenatoside.Methods:Under the simulated physiological condition,utilize fluorescence quenching and synchronization fluorescence spectrum techniques to detect the quenching process on BSA with the increasing concentration of crenatoside at different temperatures.Results and Conclusion:Crenatoside has a stronger quenching effect on BSA,which belongs to static quenching.At normal temperature,the quenching constant of crenatoside and BSA is 5.14×10 12 L·mol-1;the binding constant is 2.95×10 5 L·mol-1;the binding-site number is 1.17.The interaction type between crenatoside and BSA is speculated according to thermodynamic data;the result of synchronization fluorescence spectrum indicates that crenatoside has a less effect on the conformation of BSA.
