摘要
目的探讨谷氨酰胺联合吉西他滨对人肺腺癌A549细胞增殖、凋亡及X-连锁凋亡抑制蛋白(XIAP)表达的影响。方法体外培养人肺腺癌A549细胞,将人肺腺癌A549细胞株分别经空白对照(无药物干预)、吉西他滨(0.01 mg/L)、谷氨酰胺(32 mmol/L)、联合用药[(谷氨酰胺(32 mmol/L)联合吉西他滨(0.01 mg/L)]作用24、48、72 h后采用CCK-8法检测各组人肺腺癌细胞不同时间点增殖抑制率变化情况,使用流式细胞仪检测细胞凋亡率,应用酶联免疫吸附试验检测XIAP蛋白表达的变化。结果联合用药组人肺腺癌A549细胞增殖抑制率明显高于其他各组,并随着作用时间的延长,增殖抑制作用增强,差异均有统计学意义(P<0.05);联合用药组人肺腺癌A549细胞凋亡率明显高于其他各组,差异均有统计学意义(P<0.05)。与空白对照组比较,吉西他滨组人肺腺癌A549细胞XIAP表达明显升高,谷氨酰胺组、联合用药组人肺腺癌A549细胞XIAP表达均明显降低,差异均有统计学意义(P<0.05)。结论谷氨酰胺联合吉西他滨能显著增强吉西他滨对人肺腺癌A549细胞生长的抑制作用,该作用可能与通过下调XIAP表达从而增强肺癌细胞对吉西他滨的敏感性有关。
Objective To investigate the effects of glutamine(GLN)combined with gemcitabine on proliferation,apop-tosis and XIAP expression of human lung adenocarcinoma A549 cells.Methods A549 cells were cultured in vitro.After acting on A549 cells for 24,48,72 h by the blank control(without medication intervention),gemcitabine(0.01 mg/L),GLN(32 mmol/L)and combined medication(GLN 32 mmol/L combined with gemcitabine 0.01 mg/L),the proliferation inhibition rate of A549 cells at different time points was detectedby(CK-8),the cellular apoptosis rate was detected by using flow cytometry and the expression of XIAP protein was detected by ELISA.Results The cell proliferation inhibition rate of the combination group was obviously higher than that of the other groups,and the proliferation inhibition effect became stronger with the action time extending,the differences were statistically significant(P<0.05);the A549 cell apoptosis rate in the combined medication group was significantly higher than that in the other groups,the differences were statistically significant(P<0.05).Compared with the blank control group,the expression of A549 cell XIAP in the gemcitabine group was significantly increased,while which in the GLN group and combined medication group was significantly decreased,the differences were statistically significant(P<0.05).Conclusion GLN combined with gemcitabine can strongly enhance the growth inhibiting effect on human lung cancer A549 cells,its action may be related with down-regulating XIAP expression,thus enhancing the sensitivity of A549 cells to gemcitabine.
