HER-2阳性乳腺癌生物学特征与分子靶向治疗研究进展 被引量:9

摘要 乳腺癌是一种严重危害女性身心健康的恶性肿瘤,全球每年约有135万女性患乳腺癌,根据不同基因的表达程度将乳腺癌分为4种亚型,其中1类为HER-2受体(human epidermal growth factor receptor 2)阳性乳腺癌,约占乳腺癌总数的25%[1]。HER-2又称人表皮生长因子受体2,负责传导信号,促进细胞生长分裂[2]。研究表明,HER-2的表达水平与肿瘤的分级、大小,淋巴结转移及预后密切相关,而HER-2高表达往往预示着乳腺癌浸润性更强,对化疗药物和内分泌治疗药物不敏感,肿瘤血管增生增多,肿瘤细胞凋亡作用受到抑制等,因此HER-2的表达状况是全身治疗时药物选择的主要参考标志,也是判断乳腺癌预后不良的重要指标[2-5]。据报道,HER-2高表达与肿瘤恶性表型和耐药密切相关,且HER-2蛋白位置表浅易被抗体接近,故HER-2酪氨酸激酶受体是HER-2高表达肿瘤的有效治疗靶点[6]。
