机械压应力对人牙龈成纤维细胞表达钙网蛋白的影响 被引量:2

摘要 目的:探究机械压应力作用下不同时间人牙龈成纤维细胞(HGFs)钙网蛋白(CALR)的表达。方法:采用组织块培养法培养HGFs,将细胞接种在PLGA膜上,采用重物加载方式对其施加25g/cm^2的压应力,按加力时间不同分为0h组(对照组)、6h组、24h组、48h组、72h组,分别采用荧光定量PCR法(qPCR)和western blotting法检测各组CALR基因和蛋白表达量。结果:随着加力时间的延长,CALR基因和蛋白表达量升高,在24h时升至最高,随后逐渐降低。结论:机械压应力可使HGFs中CALR表达上调,且随着加力时间的延长CALR表达水平增高,到达峰值后下降。 Objective:To investigate the expression of calreticulin(CALR)in human gingival fibroblasts(HGFs)at different time under the action of mechanical pressure.Methods:HGFs were cultured by tissue block culture method.The cells were seeded on the PLGA scaffold?and then subjected to 25 g/cm2 loading force for 0 h(control),6 h,24 h,48 h and 72 h,respectively.The expressions of CALR mRNAand protein were detected by fluorescence quantitative PCR(qPCR)and western blotting,respectively.Results:The mRNA and protein expressions of CALR were up-regulated with the increasing of loading duration,and reached the highest level at 24 h,then decreased gradually.Conclusion:Mechanical compressive stress could up-regulate CALR expression in HGFs.The CALR level increased with prolonged duration of loading force,and reached the peak then decreased.
