摘要
目的研究广藿香对耐药哈维氏弧菌生物膜形成的抑制作用及其对成膜相关基因表达的影响等,尝试从分子水平阐释其抑制菌膜形成的机理.方法以耐药哈维氏弧菌X140701为实验菌株,通过二倍稀释法测定广藿香对菌株的最低抑菌浓度(MIC);采用结晶紫染色法测定生物膜形成量;采用qRT-PCR检测广藿香对生物膜相关基因表达的影响.结果广藿香对X140701 MIC为31.25 mg/m L(以生药计).0.782 mg/m L广藿香水提物对X140701生物膜抑制率达37.6%;广藿香水提物可使X140701生物膜相关基因lux R、fla B基因表达量上调,lux S、hfq、omp W基因表达量下调.结论广藿香能有效抑制耐药哈维氏弧菌生物膜的形成,其抑制作用与5个成膜相关基因中的表达密切相关.
Objective To study the effect of Pogostemon cablin(Blanco)Benth.(PCB)on biofilm formation and biofilm-related gene expression of multidrug resistant Vibrio harveyi and investigate the potential molecular mechanism.Methods Multidrug resistant V.harveyi strain X140701 was used as the experimental strain.The minimum inhibitory concentration(MIC)of PCB was determined by double dilution method.The biofilm formation was examined by crystal violet assay.The expression of biofilm-related genes was detected by qRT-PCR.Results The MIC of PCB was 31.25 mg/mL.The inhibitory rate of 0.782 mg/mL PCB was 37.62%.The up-regulated genes included lux R and fla B and down-regulated genes included lux S,hfq and omp W after PCB treatment.Conclusion PCB may effectively inhibit biofilm formation of multidrug resistant Vibrio harveyi.The inhibitory effect is closely related to modulation of the expression of five biofilm-related genes.
