益气活血法对脑缺血再灌注大鼠血脑屏障通透性及t-PA/PAI的影响 被引量:1

The Effect of Yiqi Huoxue method on the permeability of blood-brain barrier and t-PA/PAI in rats with cerebral ischemia-reperfusion

摘要 目的:研究益气活血法对气虚血瘀脑缺血再灌注大鼠血脑屏障(blood brain barrier,BBB)通透性及组织型纤溶酶原激活物(tissue plasminogen activator,t-PA)和纤溶酶原激活物抑制剂(plasminogen activator inhibitor,PAI)的影响。方法:健康Wistar雄性大鼠160只,随机分为5组(模型组、假手术组、活血组、益气组和益气活血组),每组36只,通过饥饿、疲劳和高脂饮食方法制作大鼠气虚血瘀模型,然后用线栓法阻断大鼠大脑中动脉(middle cerebral artery,MCA)2 h,再灌注3 d、7 d后,测定脑水肿和伊文氏蓝(EB)含量,用免疫组化法测定脑组织t-PA和PAI蛋白表达的变化。结果:相对于模型组,益气活血组能降低脑含水量和EB含量(P<0.05),各组能减少t-PA蛋白的表达(P<0.05),增加PAI-1蛋白表达,恢复t-PA/PAI平衡。且益气活血组优于益气组、活血组。结论:益气活血法能降低BBB通透性,其机制可能是抑制t-PA蛋白表达和增加PAI-1蛋白表达而发挥保护BBB的作用。 Objective:To observe the effect of Yiqi Huoxue Method on the blood brain barrier(BBB),tissue-type plasminogen activator(t-PA),plasminogen activator inhibitor(PAI)in cerebral ischemia-reperfusion rats with Qi deficiency and blood stasis.Methods:The healthy Wistar male rats were randomly divided into 5 groups(Model group,sham operation group,Huoxue group,Yiqi group and Yiqi Huoxue group),36 rats in each group.The model of rats with Qi deficiency and blood stasis was made by starvation,fatigue and high fat diet.After the middle cerebral arteries(MCA)of rats had been occluded for 2 hours by line bolt method and reperfused for 3 or 7 days,then the status of encephaledema and the content of Evans blue(EB)were detected.Meanwhile,the protein expression of t-PA and PAI in brain tissue was detected by immuno-histochemistry method.Results:Compared with the model group,the cerebral water content and EB content in Yiqi Huoxue group were significantly reduced.The expression of t-PA protein was obviously reduced,the expression of PAI-1 protein was increased and the balance of t-PA/PAI was restored in the treatment groups.The results of Yiqi Huoxue group were better than that of Yiqi group or Huoxue group.Conclusion:The method of Yiqi Huoxue could decrease the permeability of blood-brain barrier,and its mechanism plays the role of protecting BBB probably in inhibiting the expression of t-PA protein and increasing the expression of PAI-1 protein.
