摘要
目的分析依达拉奉对糖皮质激素在慢性阻塞性肺疾病(COPD)大鼠上抗炎作用的影响。方法选取50只实验用健康雄性SD大鼠,对其中40只进行COPD造模,造模成功后按随机数表法分为4组,分别为COPD组、布地奈德组、依达拉奉组及联合干预组(依达拉奉+布地奈德),各10只,未造模的10只SD大鼠作为空白组。测定各组大鼠肺功能,留取血液标本、肺泡灌洗液,测定血液及肺泡灌洗液中白细胞及中性粒细胞计数。结果 COPD组FEV_(0.3)/FVC%、PEF水平均低于空白组,差异有统计学意义(P均<0.05);布地奈德组、依达拉奉组的FEV_(0.3)/FVC%、PEF均高于COPD组,差异有统计学意义(P均<0.05);联合干预组的FEV_(0.3)/FVC%、PEF均高于COPD组、布地奈德组和依达拉奉组,差异有统计学意义(P均<0.05)。COPD组动脉血及肺泡灌洗液中的白细胞、中性粒细胞计数均高于空白组,差异有统计学意义(均P<0.05);布地奈德组、依达拉奉组动脉血及肺泡灌洗液白细胞、中性粒细胞计数均低于COPD组,差异无统计学意义(P均>0.05);联合干预组动脉血及肺泡灌洗液中白细胞、中性粒细胞计数均低于COPD组、布地奈德组、依达拉奉组,差异有统计学意义(P均<0.05)。结论依达拉奉联合糖皮质激素能够在一定程度上增强抗炎效果。
Objective To explore the effect of edaravone on anti-inflammatory of glucocorticoid in chronic obstructive pulmonary disease(COPD)rats.Methods Forty healthy male SD Rats were randomly divided into healthy blank control group(n=10)and model group(n=40)(including COPD group,budesonide group,edaravone group and combined intervention group,10 rats in each group).Pulmonary function,neutrophil and leukocyte levels in serum and alveolar lavage fluid were measured.Results The levels of FEV 0.3/FVC%and PEF were lower in COPD group than those in control group(P<0.05).The levels of FEV 0.3/FVC%and PEF were higher in budesonide group and edaravone group than those in COPD group(P<0.05).The levels of FEV 0.3/FVC%and PEF in combined intervention group were significantly higher than those in COPD group,budesonide group,edaravone group,and the differences were statistically significant(P<0.05).The percentages of leukocyte and neutrophils in the arterial and alveolar lavage fluid of COPD group were significantly higher than those of the blank group(P<0.05).The leukocyte and neutrophil counts of arterial blood and alveolar lavage fluid in budesonide group and edaravone group were lower than those in COPD group,but the differences were not statistically significant(P>0.05).The percentage of leukocyte and neutrophils in the arterial blood and alveolar lavage fluid of the intervention group were significantly lower than those of the COPD group,budesonide and edaravone group,and the differences were statistically significant(P<0.05).Conclusion Edaravone combined with budesonide can partly improve the effect of anti-inflammatory of glucocorticoid.
