miR-25在乳腺癌组织、细胞的表达及对细胞增殖的影响 被引量:4

摘要 目的探讨miR-25在乳腺癌组织和细胞系中表达及对细胞增殖的影响。方法采用实时荧光定量PCR法检测miR-25在乳腺癌组织、癌旁组织及乳腺癌细胞系SK-BR-3、MCF-7、MDA-MB-231和人乳腺上皮细胞系HBL-100中的表达,分析miR-25表达与患者临床病理特征的关系。分别将miR-25 inhibitor和inhibitor-NC转染至SK-BR-3细胞,转染后的SK-BR-3细胞分为miR-25 inhibitor组和inhibitor-NC组,通过CCK-8试验检测乳腺癌细胞增殖能力。结果 miR-25在乳腺癌组织中的表达高于癌旁组织(P<0.01),miR-25在各乳腺癌细胞系中的表达高于正常乳腺上皮细胞系(P均<0.01)。乳腺癌组织中miR-25表达与TNM分期、淋巴结转移有关(P均<0.01),而与年龄、肿瘤最大直径、ER、PR、C-erb B-2及组织学分级等无关(P均>0.05)。miR-25 inhibitor组和inhibitor-NC组细胞中miR-25的相对表达量分别为0.38±0.11、1.01±0.03,两组比较,P<0.01。CCK-8实验结果显示,miR-25inhibitor组SK-BR-3细胞的增殖速度较inhibitor-NC组下降(P<0.05)。结论 miR-25在乳腺癌组织和细胞系中呈高表达,下调miR-25表达能明显抑制乳腺癌细胞增殖。
