摘要
目的探讨姜黄素对百草枯致急性肺损伤小鼠肺组织丝裂原活化蛋白激酶(mitogen-activated protein kinase,MAPK)通路蛋白表达的影响。方法 90只小鼠按随机数字表法分为3组,每组30只,分别为:对照组、百草枯模型组、姜黄素干预组。对照组给予腹腔注射1 m L 0.9%的Na Cl溶液中,模型组给予含15 mg/kg的百草枯的生理盐水;干预组则在造模之前15 min给予200 mg/kg的姜黄素玉米油混悬液。3组小鼠均分为6 h、24 h、72 h三个时间点,每个时间点处死10只。分别比较3组小鼠肺组织的丙二醛(MDA)含量、超氧化物歧化酶(SOD)活性、肺组织损伤评分以及MAPK信号通路蛋白的表达。结果与对照组比较,模型组给予百草枯后,小鼠肺组织的MDA含量、损伤评分在各时间点均显著升高(P<0.05),SOD活性均显著降低(P<0.05),而姜黄素干预后,MDA含量和损伤评分则均显著降低(P<0.05),SOD活性显著升高(P<0.05)。MAPK通路蛋白的磷酸化水平在对照组中仅有少量表达,而给予百草枯后,表达量显著升高(P<0.05),使用姜黄素干预后,各蛋白的磷酸化水平又显著降低(P<0.05)。结论 MAPK信号通路在姜黄素干预的百草枯致急性肺损伤过程中发挥重要的作用,其通过抑制MAPK磷酸化,减少百草枯引起的肺损伤,可为百草枯中毒治疗提供新方向。
Objective To investigate the effect of curcumin on expression of MAPK pathway protein in lung tissue of mice with acute lung injury induced by paraquat.Methods 90 mice were randomly and evenly divided into 3 groups,the control group,the paraquat model group,and the curcumin intervention group.The control group was given intraperitoneal injection of 1 mL 0.9%of NaCl solution,the model group was given with 15 mg/kg paraquat 0.9%of NaCl solution,and the intervention group was given 200 mg/kg of curcumin corn oil suspension 15 min before the model.Each 10 mice were killed at 6 h,24 h and 72 h after treatment.The content of malondialdehyde(MDA),superoxide dismutase(SOD)activity,lung injury score and the expression of MAPK signaling pathway proteins were compared among the 3 groups.Results Compared with the control group,the content of MDA and the lung tissue of mice score at each time point increased significantly(P<0.05),and the activity of SOD decreased significantly(P<0.05)in the paraquat model group.The effect of curcumin on MDA content and injury score decreased significantly(P<0.05),and the activity of SOD increased significantly after curcumin intervened(P<0.05).MAPK pathway protein phosphorylation in the control group were only a small amount of expression,and the expression increased significantly for paraquat addition(P<0.05).The protein phosphorylation level was significantly reduced after curcumin intervention(P<0.05).Conclusion The MAPK signaling pathway plays an important role in the curcumin intervention on acute lung injury induced by paraquat,which is through inhibition of MAPK phosphorylation to reducing the paraquat induced lung injury,and can provide a new direction for the treatment of paraquat poisoning.
