摘要
日本鳗鲡(Anguilla japonica)野生资源数量锐减,亟需对其开展群体遗传学和保护遗传学研究。单核苷酸多态性标记(SNP)是重要的遗传标记,然而在日本鳗鲡中还尚未有报道。本研究采用酶切位点相关DNA测序技术开发了日本鳗鲡的离散SNP标记,利用KASPar技术对辽宁丹东和福建三沙两个群体共61尾玻璃鳗样品进行SNP分型,验证所开发SNP标记的多态性。研究共得到38个多态性较高的离散SNP标记。在丹东群体中,这38个位点的观测杂合度(Ho)为0.000~0.407,期望杂合度(He)为0.033~0.484;在三沙群体中,这38个位点的观测杂合度(Ho)为0.032~0.500,期望杂合度(He)为0.032~0.494。这表明已处于濒危状态的日本鳗鲡依然具有较高的遗传多样性水平。丹东群体中的AJ_SNP_03位点和三沙群体中的AJ_SNP_25和AJ_SNP_35位点偏离哈温平衡,所有位点间均不存在连锁不平衡现象。丹东群体和三沙群体间的遗传距离FST值为-0.005(P=0.898),表明这两个群体间无显著的遗传分化,符合日本鳗鲡随机交配的属性。研究结果表明,本研究开发的SNP标记可以用于解析日本鳗鲡的群体遗传结构及探究其本地适应性,为其资源的保护提供基础资料。
Japanese eel(Anguilla japonica)has drastically declined in past three decades and requires conservation.SNPs are important molecular markers for the study of conservation genetics,while they are still absent for this species.In the present study,we developed outlier SNP markers for Japanese eel using RAD-Seq(restriction site-associated DNA sequencing).The polymorphism of the markers was validated by performing KASPar(KBiosciences Competitive Allele-Specific PCR genotyping system)genotyping of 61 specimens from Dandong,Liaoning Province and Sansha,Fujian Province.A total of 38 polymorphic SNPs were developed.Within Dandong(DD)population,observed and expected heterozygosity across 38 loci ranged from 0.000 to 0.407 and from 0.033 to 0.484,respectively.Within Sansha(SS)population,observed and expected heterozygosity across 38 loci ranged from 0.032 to 0.500 and from 0.032 to 0.494,respectively.Locus AJ_SNP_03 in Dandong and loci AJ_SNP_25 and AJ_SNP_35 in Sansha showed a significant departure from HWE after Bonferroni correction.No linkage disequilibrium was detected within two populations.The FST between two populations was-0.005,not statistically significant(P=0.898),which was consistent with the panmictic hypothesis.These markers may aid to studying the genetic population structure and local adaptation of Japanese eel,vitally important for the management and protection of eel resource.
