摘要
该研究建立了一种利用高效液相色谱法测定黑曲霉(Aspergillus niger)xj发酵液中5-羟甲基糠醛(5-HMF)含量的检测方法。样品经乙醚提取,采用Spheriorb C18柱(4.6 mm×150 mm,5μm)为色谱柱,以乙睛:0.1%乙酸水(冰乙酸调)=10∶90(V/V)为流动相,柱温30℃;流速1.0 mL/min,检测波长为□=277 nm。在该色谱条件下,5-HMF呈现较好的分离效果和重现性,其含量与峰面积呈现良好线性关系(R为0.999),保留时间2.436 min,回收率87.12%~90.78%,精密度实验结果相对标准偏差(RSD)0.71%。测得发酵液样品中5-HMF含量为0.125 7 mg/g。该方法准确可靠,重复性好,可对黑曲霉发酵液中5-HMF的含量测定作参考作用。
In this study,a method for determination of 5-hydroxymethyl-furfural(5-HMF)in fermentation broth of Aspergillus niger xj was established by HPLC.The samples were extracted with diethyl ether,with Spheriorb Ci8(4.6 mmx150 mm,5^m)as chromatographic column,with acetonitrile and 0.1%acetic acid water(ice acetic acid)=10-90(V/V)as mobile phase at 30 V,.The flow rate was 1.0 ml/min and the determine wavelength was 277 nm.Under the chromatographic conditions,the 5-HMF showed good separation effect and reproducibility,and its content was in good linear relationship with the peak area(R=0.999).The retention time of 5-HMF was 2.436 min and the recovery rate was 87.12%-90.78%,with relative standard deviation(RSD)0.71%of precision experiments results.The content of 5-HMF in fermentation broth sample was 0.125 7 mg/g.The method was accurate,reliable and with good reproducibility,and could be used as a reference for the determination of 5-HMF in fermentation broth of A.niger.
