血清肿瘤自身抗体检测对乳腺癌诊断价值分析 被引量:1

摘要 目的 :探讨血清中肿瘤抗原Tn、TP53和人表皮生长因子受体2(human epidermal growth factor receptor-2,HER2)自身抗体水平与乳腺癌发病和发展的相关性。方法 :采用病例对照研究设计,选取30例乳腺癌患者和19例正常人为研究对象。采用酶联免疫吸附法(enzyme-linked immunoabsorbent assay,ELISA)检测肿瘤糖基抗原Tn、TP53和HER2自身抗体的水平,通过受试者工作特征曲线(receiver operating characteristic curve,ROC曲线)和Mann-Whitney检验研究3个肿瘤自身抗体指标与乳腺癌发病风险以及癌症分期的相关性。结果:ELISA检测发现乳腺癌患者血清中抗Tn自身抗体高于正常人群2倍以上,抗TP53和HER2自身抗体显著升高,而正常人群血清样本中几乎检测不到,且3种自身抗体与癌症分期呈负相关。结论:抗糖基抗原Tn、TP53和HER2自身抗体有望成为乳腺癌早期诊断和治疗效果评判检测指标。 Objective:To assess the relationship between tumor antigen[Tn,TP53 and human epidermal growth factor receptor-2(HER2)]autoantibody level and breast cancer occurrence and development.Methods:A case-control study among 30 breast cancer cases and 19 cases controls matched by age was conducted.Three breast cancer related tumor antigen autoantibodies were selected as new biomarker candidates.Taking advantage of tumor antigen-coated enzyme-linked immunoabsorbent assay(ELISA),all patient serum samples were detected to evaluate each autoantibody level and statically analyzed with Mann-Whitney test and receiver operating characteristic curve(ROC curve)to evaluate the correlation between tumor autoantibody level and breast cancer occurrence and stages.Results:ELISA results showed that serum Tn autoantibody level of breast cancer patient increased two times more compared with health candidates and autoantibody level is reversely correlated with cancer stage;although TP53 and HER2 autoantibody level of health candidates were almost undetectable,their autoantibody level significantly increased in breast cancer patients.In addition,these three autoantibody level was reversely correlated with cancer stage.Conclusion:Anti-Tn,TP53 and HER2 autoantibody biomarkers can distinguish breast cancer group from control group.These three biomarker panel might have potential in breast cancer early-diagnosis and treatment-efficacy evaluation.
