摘要
目的比较新疆假龙胆不同极性部位及不同部位中獐牙菜苦苷、龙胆苦苷和獐牙菜苷的含量。方法新疆假龙胆全草乙醇提取物,依次用石油醚、乙酸乙酯和正丁醇萃取,得到不同极性部位。采用HPLC法,以甲醇-4mL·L-1磷酸水溶液(19∶81)为流动相(流速1.0mL·min-1)同时测定新疆假龙胆不同极性部位及不同部位中獐牙菜苦苷、龙胆苦苷和獐牙菜苷的含量。结果獐牙菜苦苷、龙胆苦苷和獐牙菜苷质量浓度分别在4.04~129.54(r=0.999 4),3.96~126.73(r=0.999 3)和0.93~29.70mg·L-1(r=0.999 5)的范围内线性关系良好。3种化合物含量在正丁醇部位中最高,在乙酸乙酯和水部位中较少,在石油醚部位基本检测不到;3种化学成分含量在不同部位中存在明显差异,含量顺序为花>叶>根>茎。结论花、叶和根具有较高的药用价值,醇提物经萃取后,环烯醚萜类成分得到有效的富集;该方法专属性强、准确度高、重复性好,可用于比较新疆假龙胆药材不同极性部位及其不同部位活性成分的分布情况,为后续合理开发利用新疆假龙胆提供依据。
Objective The contents of swertiamarin,gentiopicroside and sweroside were compared in different polar fractions and different parts of Gentianella turkestanorum.Methods The ethanol extract of Gentianella turkestanorum was extracted with petroleum ether,ethyl acetate and n-butyl alcohol,successively to obtain different polarity fractions.By using methanol-4mL·L-1phosphoric acid solution(19∶81)as the mobile phase(at a flow rate of1.0mL·min-1),swertiamarin,gentiopicroside and sweroside contents were simultaneously determined by HPLC.Results The good linear relationships were found in the ranges of4.04-129.54(r=0.9994),3.96-126.73(r=0.9993)and0.93-29.70mg·L-1(r=0.9995)for swertiamarin,gentiopicroside and sweroside,respectively.The contents of the3compounds were the highest in n-butanol fraction,less in ethyl acetate and water fractions,and no detectable in the petroleum ether fraction.The contents of3compounds showed significant difference in different parts of Gentianella turkestanorum,flower>leaf>root>stem.Conclusion The method is specific,accurate and reproducible.It can be used to compare the distribution of the active ingredients in Gentianella turkestanorum herbs and different polar fractions of the alcohol extract,and also provide an experimental evidence for the subsequent research of Gentianella turkestanorum pharmacological active fractions.
