摘要 目的探讨基质金属蛋白酶(matrix metalloproteinases,MMP)在口腔鳞状细胞癌(oral squamous cell carci noma,OSCC)中的表达及对其生物学行为的影响。方法免疫组化检测口腔癌和正常口腔组织中M M P-2和M M P-9的表达情况;分析高表达MMP-2和MMP-9和临床颈淋巴结转移之间的关联;划痕实验检测MMP-2和MMP-9的表达对口腔鳞状细胞癌细胞迁移能力的影响;Transwell试验检测MMP-2和MMP-9的表达对口腔鳞状细胞癌细胞侵袭能力的影响。结果与正常口腔组织相比,口腔癌组织中MMP-2和MMP-9的表达明显上调;MMP-2和MMP-9的高表达与临床患者颈淋巴结转移有正相关关系;抑制MMP-2和MMP-9的表达可以抑制对口腔鳞状细胞癌细胞的迁移侵袭能力。结论 MMP-2和MMP-9在口腔癌中表达明显升高,抑制MMP-2和MMP-9的表达可以调控口腔鳞状细胞癌的生物学行为。 Objective To investigate the expression of matrix metalloproteinases(MMPs)in oral squamous cell carcinoma and their biological behavior.Methods The expressions of MMP-2and MMP-9in oral and normal oral tissues were detected by immunohistochemistry.The association between high expressions of MMP-2and MMP-9and clinical cervical lymph node metastasis was analyzed.The effect of MMP-2and MMP-9expressions on the migration of oral squamous cell carcinoma cells was examined by scratch test.The effect of MMP-2and MMP-9expressions on the invasive ability of oral squamous cell carcinoma cells was detected by Transwell assay.Results Compared with normal oral tissue,the expressions of MMP-2and MMP-9in oral squamous cell carcinoma were significantly up-regulated.The high expressions of MMP-2and MMP-9were positively correlated with cervical lymph node metastasis in clinical patients.Inhibition of MMP2and MMP-9expressions can inhibit the migration and invasion of oral squamous cell carcinoma cells.Conclusions The expressions of MMP-2and MMP-9in oral cancer are significantly increased.Inhibition of MMP-2and MMP-9expressions can regulate the biological behavior of oral squamous cell carcinoma.
