摘要
目的探讨猴头菌多糖(HPS)对Lewis肺癌荷瘤小鼠肿瘤抑制作用及其可能机制。方法选取小鼠50只均接种Lewis肺癌瘤株造模,随机分为HPS高、中、低剂量组、环磷酰胺(CTX)组和模型对照组,每组10只,HPS不同剂量组分别给予HPS 200 mg/(kg·d)、100 mg/(kg·d)、50 mg/(kg·d),连续给药10 d;CTX组给予CTX 40 mg/(kg·d),连续给药3 d;模型对照组每天给予生理盐水0.1 mL/10 g。另选取10只健康小鼠作为正常对照组。10 d后对各组小鼠眼球取血,分离血清,ELISA法测定肿瘤坏死因子-α(TNF-α)和白细胞介素-2(IL-2),分离瘤体,称质量并计算抑瘤率。结果与模型对照组相比,CTX组和HPS高、中、低剂量组的瘤重均显著降低(P<0.05),抑瘤率分别为73.52%、44.33%、29.65%和16.58%,各组间差异均有统计学意义(P<0.05)。与正常对照组相比,模型对照组小鼠TNF-α显著增高,IL-2显著降低(P<0.05);与模型对照组相比,CTX组和HPS各剂量组的TNF-α均显著降低,IL-2均显著增高(P<0.05),HPS各剂量组变化幅度显著小于CTX组(P<0.05),但HPS各剂量组间无统计学意义(P>0.05)。结论 HPS对小鼠Lewis肺癌具有剂量依赖性的抑瘤作用,其机制可能与改善血清TNF-α和IL-2水平,进而改善机体免疫功能有关。
Objective To study the inhibition and its possible mechanisms of Hericium polysaccharides(HPS)on mice with Lewis lung cancer.Methods50mice were chosen to establish Lewis lung cancer mice models,and were divided randomly into five groups:HPS high-dose group,HPS middle-dose group,HPS low-dose group,cyclophosphamide(CTX)group and model control group,10mice in each group.The mouse in former three groups were given HPS200mg/kg,100mg/kg and50mg/kg daily for10days,respectively,while those in CTX group were given CTX40mg/kg daily for3days,and those in model control group were given physiological saline0.1mg/10g daily for10days.Another10healthy mice were chosen as the normal control group.The blood from eyeball of mice were collected,and the blood serum were separated,and tumor necrosis factor-α(TNF-α)and interleukin-2(IL-2)were detected by ELISA.The tumor inhabitation rates were calculated.Results Compared with model control group,the tumor weights in CTX group and HPS high-dose,middle-dose and low-dose group all significantly decreased(P<0.05)with tumor inhabitation rates73.52%,44.33%,29.65%and16.58%,which had statistical significance between each group(P<0.05).The level of TNF-αwas higher and the level of IL-2in model control group was lower than those in normal control group(P<0.05).Compared with model control group,the level of TNF-αsignificantly increased and the level of IL-2significantly decreased in CTX group and HPS groups(P<0.05).The change trends of CTX group were more obvious than those in HPS groups(P<0.05),while there was no significant differences among HPS different dose groups(P>0.05).Conclusion HPS has dose-dependent inhibitory effect on mice with Lewis lung cancer,which may be associated with improvement of serum TNF-αand IL-2level,thus improving immune function.
