摘要
建立小鼠肝组织磷脂提取及薄层层析(TLC)分析方法,实现对小鼠肝脏磷脂各组分比例的快速测定。通过对磷脂提取过程中氯仿―甲醇混合抽提液比例的优化得到氯仿―甲醇的体积比为1:2时可有效提取磷脂;通过优化TLC展开剂得到氯仿―甲醇―水体积比为65∶25∶4时磷脂各组分可有效分离。可初步得出PC、PE的相对含量,为后续实验是否采用气相或高效液相色谱准确定量分析磷脂各组分含量做初步判断。
To determine the proportion of phospholipids in mice liver rapidly,the method of phospholipid extraction and thin layer chromatography of liver tissue in mice was established.By optimizing the proportion of chloroform-methanol mixed extraction liquid,the volume ratio of Chloroform to methanol is1∶2which can extract phospholipid efficiently.Through optimizing of thin layer chromatography,when Chloroform:methanol:water ratio is65∶25∶4,the components[JP]of phospholipid can be separated effectively.This method arrives at the relative content of Phosphatidylcholine and Phosphatidylethanolamine preliminarily,which judges for determining phospholipid extraction using gas chromatography or high performance liquid chromatography accurately.
