摘要
选用健康的SD雌性大鼠,在发情期或发情间期与雄鼠交配,在雌鼠受孕后的6~15d灌胃10、33.3和100mg/kg的硫磺负离子添加剂混合液,对照组为1%羧甲基纤维素(CMC)溶液,至受孕第20天处死动物,检测母体体重、摄食量,观察孕鼠的临床症状及剖检后有无内脏病变等情况;检查胎盘重量、黄体数、着床率、活胎率、胎儿的重量、体长、内脏、骨骼发育情况、雌雄比等,对硫磺负离子添加剂的致畸敏感期生殖毒性进行综合评价。结果表明:SD雌性大鼠口服硫磺负离子添加剂混合液10、33.3、100mg/kg后,受孕雌鼠的各项测定指标和胎儿内脏及骨骼检查等各项检测指标与对照组比较,差异无统计学意义(P>0.05),母体无明显毒性反应,胎儿也没出现明显的致畸作用。硫磺负离子添加剂在本试验所用剂量时无毒性和致畸作用,未观察到有害作用剂量(NOAEL)不小于100mg/kg。
Sulfur anion additives were evaluated in developmental toxicity studies with SD rats in this research.The pregnant SD rats were gavaged with0(vehicle control),10,33.3and100mg/kg respectivelty after6~15d from gestation day(GD).After20d,the rats were killed and the weight,food intake,the clinical symptoms and visceral lesions of female SD rats were observed,placental weight,numbers of corpus luteum,the rates of implantation and live birth,the weight,length,the development of organs and bones for fetus as well as the sex ratio were also detected for the comprehensive evaluation of reproductive toxicity for this additive.The results showed that difference between treatment groups and control group,has no statistics significance(P>0.05),which proves the dose of sulfur anion additives used in this experiment have no toxic and teratogenic effect for pregnant SD rats,and the no observed adverse effect level(NOAEL)for maternal and developmental toxicity in SD rats was the highest dose tested of100mg/kg bw/day.
