氮素对水稻幼苗氮代谢相关酶活性及相关基因表达的影响 被引量:2

摘要 为明确氮素对不同蛋白质含量水稻幼苗的影响,用营养液培养的方法,对4个稻米蛋白质含量有差异的粳稻品种幼苗进行诱导处理,研究了水稻幼苗在0.05mol·L^(-1) KNO3处理下,各品种水稻干物质积累的差异,以及硝酸还原酶(NR)、谷氨酰胺合成酶(GS)活性和NR基因(OsNR1)、GS基因(OsGs1^(-1))表达差异。结果表明:经0.05mol·L^(-1) KNO3处理后各品种地上部和根干重均显著高于对照,叶片和根中硝态氮(NO_3^--N)和铵态氮(NH_4^+-N)含量显著高于对照,叶片中NR和GS活性显著高于对照;随着氮素处理时间的增加,OsNR1基因相对表达量增加,在处理2h时达到最大值,而后呈随处理时间的增加呈下降的趋势;在氮素处理前期,OsGs1^(-1)的表达量变化不大,而在处理8h时,OsGs1^(-1)基因的表达量显著增加,此时的基因相对表达量是处理前的1.2倍。 In order to determine the effect of nitrogen on rice seedlings of different protein content,4japonica rice seedlings with different contents of protein treated were induced by hydroponics.The differences of dry matter accumulation,nitrate reductase(NR)and glutamine synthetase(GS)activities and the expression of OsNR1,OsGs1-1genes were studied under0.05mol·L-1KNO3treatments on japonica rice seedlings with4different protein content.The results showed that after induced by0.05mol·L-1KNO3,the dry weights of over ground part and root were higher than control,the contents of NO-3-N and NH+4-N in leaves and roots were significantly higher than the control,NR and GS activities in leaves were significantly higher than the control,OsNR1gene expression relatively enhanced with the increase of nitrogen treatment time,the maximum value was reached when2h treated,and then decreased with time;the change in OsGs1-1expression is not not obvious,OsGs1-1gene expression significantly increased and was1.2times that before treatment when treating for8h.
