摘要
运用分子生物学技术及多元统计分析的方法,跟踪湖泛爆发过程中细菌群落的动态及环境因子变化。结果表明:湖泛爆发过程中水质明显恶化,DO(溶解氧)、SD(透明度)、p H(酸碱度)显著降低,其中DO含量均低于0.51 mg·L^(-1),水体中TN(总氮)、TP(总磷)等营养盐的含量远远高于正常水体中的含量。细菌丰度和藻类数量明显升高,其中,湖泛中心区的细菌丰度上升到107 cells·L^(-1),相比未发生湖泛的区域提高了一个数量级。运用高通量测序技术,分析比较非湖泛区及不同程度湖泛区的细菌群落结构的差异。结果表明:湖泛最严重的区域,α-、γ-变形菌纲(alpha-,gamma-Proteobacteria)分别由未发生湖泛时的4.10%和13.16%降至0.59%和1.91%;放线菌门(Actinobacteria)所占比例由未发生湖泛区域的19.04%降至0.92%;而拟杆菌门(Bacteriodetes)所占的比例则由1.12%增加至36.32%。此外,还检测到一些特殊生境的细菌类群,如Ilumatobacter,Chitinophagaceae。通过对湖泛事件中细菌群落与关键环境因子的变化规律研究,为湖泛问题的深入研究提供一定科学依据。
Molecular biology techniques and multivariate statistical analysis method were used to evaluate relationships between bacterial abundance and environmental variables in the areas when black bloom happened regularly.We also investigated the changes of spatial bacterial community during black bloom happened.Results showed that:DO(dissolved oxygen),SD(secchi disk)and pH(potential of hydrogen)decreased significantly in the black bloom field,particularly,DO decreased as low as0.51mg·L-1,which indicated an anaerobic environment in the water column.In addition,black bloom increased nutrient concentrations(total nitrogen,total phosphorus),phytoplankton biomass and bacterial abundance.The bacterial abundance in the central of black booms area exceeded107cells·L-1,which improved an order of magnitude higher than other areas without black bloom;bacterial community structures were significantly different among the areas:alpha-,beta-,and gamma-Proteobacteria,Bacteriodetes,Actinobacteria.Namely,along black bloom,α-,γ-proteobacteria decreased from4.10%and13.16%to0.59%and1.91%respectively;and Actinobacteria deceased from19.04%to0.92%.In contrast,Bacteriodetes increased from1.12%to36.32%.In addition,some special habitats bacteria were also detected in our samples like Ilumatobacter and Chitinophagaceae.Through studying the change rules of bacterial community and key environmental factors when black bloom occurs,we can provide some scientific instruction for further research of black bloom.
