摘要
目的:利用网络药理学技术预测中药复方桂枝甘草汤治疗心律失常的作用机制。方法:通过TDT数据库获取桂枝甘草汤相关活性成分的化学结构;通过HIT和TTD数据库筛选桂枝甘草汤的作用靶点,建立靶点数据集;OMIM数据库筛选心律失常相关的靶标;PPI数据库构建桂枝甘草汤和心律失常的交互靶标;用Cytoscape软件构建"成分-靶点-疾病"交互网络图。结果:筛选出桂枝甘草汤中41个活性成分和共同直接作用的45个蛋白靶点;通过度、介度中心度、接近中心度等网络拓扑特征评价筛选出与桂枝甘草汤在心律失常方面作用最相关靶标,利用KEGG数据库对入选靶标进行相关通路富集,筛选出37条通路在心律失常方面具有作用。结论:通过网络药理学预测了桂枝甘草汤中桂枝和甘草配伍治疗心律失常的体内作用机制,为其活性成分研究和临床应用提供理论依据。
Objective:To predict the mechanism of Guizhi Gancao Soup in the treatment of arrhythmia by using the technique of network pharmacology.Methods:TDT database was used to screen the chemical composition of Guizhi Gancao Soup.The HIT and TTD database was used to predict the targets of the Soup;OMIM database was applied to screen the targets of arrhythmia;PPI database established the interaction between the drug target and the disease target.The“compound-target-disease”complex network diagram was constructed by the use of the Cytoscape software.Results:We screened41active ingredients in Guizhi Gancao Soup which had a direct effect on45targets.Targets of Guizhi Gancao Soup were selected,which played an important role in the anti-arrhythmia process by using such network topological parameters as Degree,Betweenness Centrality and Closeness Centrality.In the“component-targetdisease”network of Guizhi Gancao Soup,the targets which achieve the standard were enriched by the KEGG database;37signal pathways were connected with the anti-arrhythmia effects.Conclusion:The mechanism of Guizhi Gancao Soup in the treatment of arrhythmia is predicted by the use of the network pharmacology,which provides the theoretical basis for its clinical application.
