摘要
通过微波辅助提取和热水浸提两种方法,提取怀牛膝茎叶多糖.并测定样品提取液对农杆菌、大肠杆菌的抑菌作用,同时测定其对超氧阴离子自由基和羟自由基的清除率,研究其抗氧化活性.结果表明,微波辅助提取和热水浸提法对怀牛膝茎叶多糖提取率分别为103.79mg/g、89.58mg/g.微波辅助提取液对农杆菌及大肠杆菌抑菌效果较好,抑菌圈直径分别为10mm、11mm.且样品提取液有明显的清除自由基活性,微波辅助法提取的多糖对超氧阴离子自由基的清除率为77.38%,对羟自由基的清除率为60.23%.
The polysaccharide was extracted by microwave-assisted and hot-water extraction from stems and leaves ofAchyranthes bidentata Bl..The antibacterial effects on bacterials such asAgrobacterium tumefacienandEscherichia coli were detected.At the same time,the antioxidant activity of the extracts was detected.The results showed that the extraction ratio of the polysaccharide through microwave-assisted and hot-water extraction were103.79mg/g and89.58mg/g,respectively.It was also found that microwave-assisted extracts had better inhibition effects onAgrobacterium tumefacienandEscherichia coli with10mm and11mm respectively of the inhibition zone diameter for the two bacterials.And the microwave-assisted extracts had obvious antioxidant activity with the clearance rate of77.38%for superoxide anion radical and60.23%for hydroxyl radical.
