摘要 目的建立全血中甲苯噻嗪的固相萃取/气相色谱质谱检测方法。方法血液样品用4倍体积的pH=6的磷酸缓冲盐稀释,离心,上清液经Oasis HLB固相萃取柱提取净化,GC/MS法进行定性定量分析,选用碎片离子m/z 220做定量离子。结果甲苯噻嗪在0.50~50.0μg/m L浓度范围内线性关系良好(r2=0.991),检出限为23.7ng/m L,在0.50μg/m L添加水平,回收率大于65.0%,方法日内RSD<6.37%,日间RSD<8.34%。结论本方法简单快速,选择性好,可用于实际案件中甲苯噻嗪的检测。 Objective To develop a method for determining xylazine in human whole blood by solid phase extraction/gaschromatography-mass spectrometry(GC/MS).Methods Blood sample was extracted and purified by Oasis HLB solid phaseextraction column after four-fold dilution with phosphate buffer(pH=6),then followed to be analyzed by GC/MS.The ions(m/z220)were applied for quantitative analysis.Results The limit of detection was23.7ng/mL and the linear ranges werefrom0.50μg/mL to50.0μg/mL(r2=0.991).The average recoveries for xylazine were over65.0%in the blood sample spikedwith the drug’s concentration at0.50μg/mL,and the relative standard deviations(RSD%)were less than6.37%for intra-dayand8.34%for inter-day determination.Conclusion The method is simple,suitable for the determination and confirmation ofxylazine in blood sample.
