调心方对APP/PS1双转基因AD小鼠空间学习记忆能力和海马突触传递长时程增强的影响 被引量:3

摘要 目的观察调心方对APP/PS1双转基因阿尔茨海默病(AD)小鼠空间学习记忆能力和海马突触传递长时程增强(LTP)的影响,探讨其作用机制。方法将54只3月龄APP/PS1双转基因AD小鼠随机分为模型组、调心方组和阳性药组,以同月龄同性别18只C57BL/6J野生小鼠作为正常组,各给药组给予相应药物灌胃。给药12周后,进行Morris水迷宫行为学检测和离体电生理记录。结果 Morris水迷宫测试结果显示,定位航行实验与模型组比较,调心方组逃避潜伏期时间明显缩短(P<0.01);空间探索实验与模型组比较,调心方组小鼠穿越原平台所在象限次数和在原平台所在象限停留时间明显增加(P<0.05,P<0.01)。在离体海马电生理诱导中,与模型组比较,调心方组海马兴奋性突触后电位斜率明显增强(P<0.01),海马双脉冲易化无明显差异(P>0.05)。结论调心方可有效改善APP/PS1双转基因AD小鼠空间学习记忆能力和神经突触可塑性,具有认知保护作用。 Objective To investigate the effects of Tiaoxin Formula on spatial learning and memory ability and long-term potentation(LTP)in the hippocampus of APP/PS1transgenic mice with Alzheimer disease;To discuss its mechanism of action.Methods Totally54three-month-old APP/PS1transgenic mice were randomly divided into model control group,Tiaoxin Formula group and positive control group,with18mice in each group.Another18three-month-old C57BL/6J wild mice were chosen as normal control group.All administration groups receive relevant medicine.12weeks later,Morris water maze test was used to test behavior and in vitro electrophysiology record.Results The Morris water maze test showed that in place navigation test,compared with the model control group,escape latency time in Tiaoxin Formula group was significantly reduced(P<0.01).In the space experiments,compared with the model control group,the number of crossing the platform quadrant and the time staying at the platform quadrant in Tiaoxin Formula group were significantly increased(P<0.05,P<0.01).In vitro hippocampal electrophysiological induction,compared with the model control group,fEPSP slope in the hippocampus in Tiaoxin Formula group were significantly increased(P<0.01),the PPF had no significant difference(P>0.05).Conclusion Tiaoxin Formula can improve the spatial learning and memory ability and LTP of APP/PS1transgenic mice with Alzheimer disease,thus can realize cognitive protection effects.
