广东沿海地区拟穴青蟹呼肠孤病毒和双顺反子病毒-1的分子流行病学调查 被引量:3

摘要 运用双重巢式PCR方法于2014年10月和11月份对广东省阳江市、台山市、中山市、惠州市(大亚湾地区、惠东地区)、汕尾市(东涌地区、红海湾地区)等主要拟穴青蟹(Scylla paramamosain)养殖地区的青蟹呼肠孤病毒(mud crab reovirus,MCRV)和青蟹双顺反子病毒-1(mud crab discistrovirus-1,MCDV-1)流行情况进行检测调查和序列分析。结果表明在调查的地区中,60%—100%的拟穴青蟹至少携带一种病毒。10月份阳江地区、台山地区、中山地区、汕尾东涌和汕尾红海湾地区拟穴青蟹MCRV携带率分别为71.42%、68.97%、70%、72.41%和64%,MCDV-1携带率分别为61.29%、24.14%、63.33%、13.79%和12%;各地区两种病毒双重感染率均在50%以下。11月份阳江地区、台山地区、中山地区、惠州大亚湾地区和惠州惠东地区拟穴青蟹MCRV携带率分别为50%、95.65%、100%、93.1%和82.76%,MCDV-1携带率分别为10%、100%、93.1%、37.93%和27.59%,台山和中山地区地区两种病毒双重感染率均在90%以上,惠州、汕尾地区以单独感染为主。五个地区获得的27株MCRV序列中,各个地区内获得的核酸序列同源性均在99.2%—100%之间,MCRV序列遗传进化基本无地域差异;获得35株MCDV-1序列中,各个地区内获得的核酸序列同源性均在91.4%—100%之间,MCDV-1序列遗传进化出现一定的地域差异。 This paper studied the infection of mud crab reovirus(MCRV)and mud crab discistrovirus-1(MCDV-1)by duplexnested-PCR.We analyzed the virus sequences obtained from the main farming area of mud crabs in Yangjiang,Taishan,Zhongshan,Huizhou(Daya Bay and Huidong)and Shanwei(Dongchong and Red Bay)in Guangdong Province in Octoberand November of2014.The results showed that60%-100%of the mud crab(Scylla paramamosain)carried at least one virusin these areas.The infection rates of MCRV were71.42%,68.97%,70%,72.41%and64%in Yangjiang,Taishan,Zhongshanand Shanwei(Dongchong and Red Bay)in October,respectively.The infection rates of MCDV-1were61.29%,24.14%,63.33%13.79%and12%in Yangjiang,Taishan,Zhongshan and Shanwei(Dongchong and Red Bay)in October,respectively.The duplex positive rate was under50%in all regions in October.The infection rates of MCRV were50%,95.65%,100%93.1%and82.76%in Yangjiang,Taishan,Zhongshan and Huizhou(Daya Bay and Huidong)in November,respectively.Theinfection rates of MCDV-1were10%,100%,93.1%,37.93%and27.59%in Yangjiang,Taishan,Zhongshan andHuizhou(Daya Bay and Huidong)in November,respectively.The co-infection rates of MCRV and MCDV-1were more than90%in Zhongshan and Taishan in November.Single infection was the most common in Huizhou and Shanwei in November.The BLAST results indicated that these27sequences of MCRV obtained from each region shared a nucleotide sequencehomology between99.2%and100%.Most of MCRV sequences showed no regional difference.But the sequences ofMCDV-1showed some regional differences.35sequences obtained from each region shared a nucleotide sequence homologybetween91.4%and100%.
