摘要
目的:探讨吡格列酮对高糖培养肾小球系膜细胞炎症的抑制作用。方法:肾小球系膜细胞培养至90%融合,换上无血清的DMEM培养基,并随机将细胞分为正常组、高糖模型组、吡格列酮低剂量组和高剂量组,继续培养48h后,进行检测。结果:高糖下的系膜细胞经吡格列酮药物处理后LDH、IL-6、TNF-α的含量、P38MAPK的表达量和NF-κB的活性都明显的下降。结论:吡格列酮能抑制高糖培养下肾小球系膜细胞炎症作用。
Objective:To investigate the effects of pioglitazone on inflammation of glomerular mesangial cells cultured in highglucose.Methods:Glomerular mesangial cells were cultured for up to 90% fusion,replaced with serum-free DMEM,and randomlywere divided into normal control group, high glucose group model pioglitazone low dose group and high dose group,continue tocultured for 48 h,and detected.Results:Under high glucose conditions of mesangial cell by drug pioglitazone treatment after thecontent of LDH,IL-6,TNF alpha and p38MAPK expression and NF-κ B activity were significantly decreased.Conclusion:Pioglitazone can inhibit mesangial cells cultured in high glucose medium inflammation.
