摘要 目的 观察中波紫外线 (UVB)照射对正常人外周血淋巴细胞 (PBL)凋亡及吞噬功能的影响。方法 采用 5 0、10 0、2 0 0和 4 0 0J/m2 不同剂量的UVB照射PBL ,培养 2 4h后以及 10 0和 2 0 0J/m2 剂量下培养 0、6、12和 2 4h后 ,用AO染色、琼脂糖凝胶电泳和流式细胞仪 (FCM )检测细胞凋亡 ,用葡萄球菌法观察受UVB处理后吞噬细胞的功能变化。结果 PBL受照后死亡方式之一是凋亡 ,AO染色法荧光显微镜计数凋亡细胞百分率及DNA梯状带检测结果发现 ,细胞凋亡有时间和剂量依赖性 ;AO染色法检测结果与FCM检测的凋亡细胞随剂量的加大及时间的延长而增多的结果不呈平行关系。同时PBL的吞噬功能有不同程度地受损。结论 UVB照射可诱导PBL凋亡 ,凋亡可呈现时间和剂量效应关系 。 Objective To study the effects of ultraviol et B(UVB) irradiation on apoptosis in human peripheral blood lymphocytes(PBL) in vitro. Methods PBL were exposed to UVB irradiation of 50, 100, 200 and 400 J/m 2 and cultivated for 24 h and exposed to UVB irradiation of 100 and 200 J/m 2 and incubated for 0, 6, 12 and 24 h. Apoptosis was measured using gel electrophoresis for DNA Ladder and AO fluorescent staining for percentage of apoptotic cells and flow cytometry (FCM). Results The predominant model of cell death in PBL after irradiation was apoptosis which is dependent on dose and time of irradiation. The time and dose dependent apoptosis measured by AO staining did not parallel that by FCM. Conclusion UVB irradiation may cause PBL apoptosis. The extent of apoptosis is dependent upon time and dose of UVB. Neutrophil apoptosis is associated with cell function.
