摘要
目的 :研究 β -榄香烯对Tca8113(口腔舌鳞癌细胞株 )细胞株诱导凋亡机制。 方法 :应用MTT比色法和流式细胞仪测定细胞周期的变化 ,并应用电子显微镜技术观察诱导分化后的亚细胞结构。结果 :β -榄香烯对Tca8113细胞株有明显的抑制作用。流式细胞仪分析不同的药物浓度、不同的作用时间细胞凋亡的发生均有显著意义 ,G1期细胞减少 ,S期细胞增多。亚细胞结构细胞线粒体肿胀变性 ,核固缩。结论 :β -榄香烯能抑制Tca8113细胞增殖 ,促进细胞凋亡 ,阻止S期细胞过程。
Objective:To investigate the apoptosis mechanism of β-elemene on Tca8113 cell lines.Methods:To evaluate the cell cycle changes with MTT colorimetric method and flow cytometry, and to observe the differentiated subcellular structure with Electron Microscope.Results:β-elemene obviously restrained the Tca8113 cell lines. Different drug concentration and different time had evident effect on apoptosis.The number of cells in G 1 stage was reduced, while that in S stage was increased.The mitochondrion swelled and degenerated,with karyopyknosis as well in subcellular structure.Conclusions:β-elemene can retrain the proliferation of Tca8113 cell lines,and accelerates cell apoptosis and prevent cell development in S stage.
