摘要
本文研制出一种新的多种系性PAP复合物。采用双扩、参比扩散及免疫沉淀试验证实辣根过氧化酶(HRP)、兔抗HRP及鼠单克隆抗HRP三者同存于一分子PAP复合物中。将其用于ELISA及免疫病理染色,效果与普通兔PAP及鼠单克隆PAP相近,但可通过两种不同桥抗体,与作为第一抗体的兔IgG及鼠(单克隆)IgG连接,代替兔PAP与鼠单克隆PAP进行染色,在不改变反应方式的情况下,能部分克服PAP复合物在应用中的种系限制。
A new PAP complex has been generated with polyclonal and monoclonal antiperoxidase antibodies. By using the double immunodiffusion, controlling dilfusion and immunoprecipitation test, it was demonstrated that such a PAP complex consists of several different molecules, ie: HRP, rabbit antiperoxidase antibody and mouse monoclonal antiperoxidase antibody. Similar effects were shown in ELISA and immunopathology test with such PAP, rabbit PAP and mouse monoclonal PAP complex, but the new PAP complex was able to combine with rabbit IgG and mouse IgG by two different bridge antibodies. Therefore, the new PAP complex could become a candidate to substitute for the rabbit PAP and mouse PAP complexes and partially reduce the species restriction of PAP method without any change of the reaction model.
