摘要
用 L-1 5培养液培养并传代 9个乳腺癌细胞系 ,收获细胞 ,提取胞浆、胞核蛋白质 ,Western印迹法观察 NFk B家族蛋白及其抑制物的表达 ;采用 2 0 J· m- 2 剂量紫外线照射细胞 ,继续培养细胞并提取蛋白质进行检测。结果表明 :1 9个乳腺癌细胞系的 p5 0、p65和 Ik B-α胞浆表达阳性率分别为 88.9%、5 5 .6%、77.8% ,胞核表达阳性率分别为 66.7%、44.4%、2 2 .2 % ;2紫外线照射后胞核 p5 0、p65和 Ik B-α阳性率分别为 1 0 0 %、1 0 0 %、3 3 .3 %。说明紫外线照射可引起胞核 p5 0、p65阳性率明显提高 ;NFk B家族蛋白对乳腺癌的发生。
To investigate Invest the expression of NFkB family proteins and the variation of the level of p50, p65, IkB α , the 9 breast cancer cell lines were used and exposed to the UV light. 9 breast cancer cell lines were cultured and cells harvested, protein of p50, p65 and IkB extracted from cytoplasm, nuclear separately to detect the expression of p50, p65, IkB proteins by Western blotting. The UV irradiation was performed at room temperature using UV C radiation (254 nm) at a dose of 20 J·m -2 . The result were as follows: ① The expression of p50, p65 and IkB was found in some of the cytoplasm and the positive rate is 88.9%, 55.6% and 77.8%. The positive rates reached 66.7%, 44.4% and 22.2% in nuclear of the cells. ② After the UV irradiation the positive rates in nuclear of p50, p65 and IkB were up to 100%, 100%, 33.3%. The UV irradiation caused NFkB family proteins level rising remarkably in nuclear. Both p50 and p65 played a role in the growth and developing of breast cancer cell.
