摘要
蛋白激酶Cε亚型 (PKCε)和蛋白酪氨酸激酸Lck均在心肌缺血预适应的信号转导中起着十分重要的作用 .构建PKCε和Lck与绿色荧光蛋白的融合蛋白的真核细胞表达载体 .将其共转染H2 93细胞进行表达 .用荧光显微镜观察到荧光共振能量转移现象 。
Protein kinase Cε(PKCε) and tyrosine kinase Lck play critical roles in intracellular signal transduction of ischemic preconditioning of cardiomyocytes. The eukaryotic expression vectors of fusion proteins of PKCε and Lck with green fluorescent proteins were constructed and cotransfected into H293 cells for expression. Fluorescence resonance energy transfer phenomena in the cotransfected cells were observed with epi-fluorescence microscope which evidence a direct interaction between PKCε and Lck in vivo.
