摘要
目的 探讨 CD5 5、CD5 9在日本血吸虫童虫逃避补体杀伤中的作用。 方法 将日本血吸虫童虫分别与人红细胞、绵羊红细胞及 PBS孵育 2 4 h,再暴露于补体 1h。在补体攻击后 72 h,计算童虫的成活率。将与人红细胞孵育过的童虫分别与不同抗体反应 ,第 1组与 Anti- CD5 5反应 ,第 2组与 Anti- CD5 9反应 ,第 3组同时与 Anti- CD5 5、Anti- CD5 9反应 ,然后再检测各组童虫对补体的敏感性。 结果 在补体攻击后的 72 h,与人红细胞、绵羊红细胞和 PBS孵育的童虫成活率分别为 6 6 .7%、33.9%和 32 .3% ,与人红细胞孵育的童虫成活率明显较高 (P<0 .0 1)。已经与人红细胞孵育过的童虫以不同抗体封闭后再暴露于补体 1h,以 Anti- CD5 5、Anti- CD5 9封闭和同时以 Anti- CD5 5、Anti- CD5 9封闭的童虫成活率分别为 5 2 .8%、5 4 .5 %和 30 .0 % ,以 Anti- CD5 5、Anti- CD5 9同时封闭的童虫成活率最低 (P<0 .0 1)。与人红细胞孵育过的童虫可被 FITC- Anti- CD5 5、FITC- Anti- CD5 9标记上特异的荧光。 结论 CD5 5、CD5 9分子与日本血吸虫童虫逃避补体杀伤有密切的关系。
Objective To study the role of CD55?CD59 in the evasion of Schistosomula japonicum from the complement mediated killing. Methods S. japonicum was incubated with human erythrocytes, sheep erythrocytes and phosphoric buffer solution (PBS) respectively for 24 hours, then exposed to complements for 1 hour, calculated the survival rates of Schistosomula 72 hours later. The Schistosomula incubated with human erythrocytes was divided into three groups again, one was reacted with Anti CD55, the other with Anti CD59, the third with Anti CD55 and Anti CD59 simultaneously, then examined the sensitivity of the Schistosomula to complement mediated killing was detected. Results The Schistosomula incubated with human erythrocytes can be labeled with fluorescence by FITC Anti CD55 as well as FITC Anti CD59, that with sheep erythrocytes and PBS can not be labeled. 72 hours later, the survival rate of Schistosomula incubated with human erythrocytes was 66.7%, that with sheep erythrocytes and PBS was 33.9%, 32.3 % respectively, that with human erythrocytes was significant higher ( P <0.01). The survival rates of Schistosomula incubated with human erythrocytes decreased in different degree after being reacted with different antibodies, that reacted with Anti CD55, Anti CD59 and with Anti CD55, Anti CD59 simultaneously was 52.8%, 54.5% and 30.0% respectively, that reacted with Anti CD55, Anti CD59 simultaneously was lowest( P <0.01). Conclusion CD55 , CD59 may play an important role in the evasion of S. japonicum from the complement mediated killing.
