摘要
目的 :研究板蓝根有效成份对低密度脂蛋白 (LPS)刺激组织内moesinmRNA表达的影响。方法 :用不同剂量的板蓝根给予小鼠灌胃 0 .5h后 ,尾静脉注射LPS ,9h后运用原位杂交法观察各组肝、肾、脾内moesinmRNA表达 ,并检测血中TNF α,NO水平。结果 :当板蓝根液为 0 .8,0 .4mL时 ,moesin表达计分分别为 (2 1.4± 18.4) ,(2 9.1± 2 2 .2 ) ,明显低于只用LPS组(113.8± 31.4) (P <0 .0 1)。当剂量达 0 .2mL时 ,表达抑制程度不大 (85 .8± 2 6 .7)。moesinmRNA表达计分值与同期所测的TNF α ,NO水平呈正相关。结论
OBJECTIVE To observe the effect of Radix-Isaditis active component on moesin mRNA expression in tissues of mice injected with LPS.METHODS Different doses of Radix-Isaditis were given orally to mice,half hour later, LPS was challenged to BALB/c mice by intravenous injection, after 9 hours, moesin mRNA expression in liver, kidney, spleen was observed by in situ hybridization. In the meantime, ELISA was used to detect serum TNF-α, Griess assay for NO levels.RESULTS When the dose of Radix-Isaditis was 0.8, 0.4 mL, the moesin expression scores were ( 21.4± 18.4),( 29.1± 22.2),respectively, lower obviously than that of the group of using LPS only( 113.8± 31.4),(P< 0.01).When the dose was 0.2 mL,the score decreased slightly( 85.8± 26.7).There was a positive correlation between moesin expression scores and serum TNF-α, NO levels(P< 0.01).CONCLUSIONS LPS may influence the moesin expression in tissues. Moesin is thought to play an important role in the LPS-mediated diseases.
