摘要
用苜蓿丫纹夜蛾核型多角体病毒（ＡｃＭＮＰＶ）ｐ７４基因的部分片段标记探针，通过ＳｏｕｔｈｅｒｎＢｌｏｔｉｎｇ将斜纹夜蛾核型多角体病毒（ＳｌＭＮＰＶ）的ｐ７４基因定位于ＸｈｏⅠ４９ｋｂ、ＥｃｏＲⅠ４４ｋｂ、ＢａｍＨⅠ３０ｋｂ片段上．通过酶切图谱分析和部分序列测定，将其ｐ７４基因精确定位．用计算机软件ＤＮＡＳＩＳ和ＰＲＯＳＩＳ对这部分ＤＮＡ序列进行分析发现其与ＡｃＭＮＰＶｐ７４基因的同源性为６９％，其可能编码多肽的氨基酸顺序与ＡｃＭＮＰＶｐ７４相应部分的同源性为６６％．
The p 74 gene of Sl MNPV was located at Eco RⅠ 4 4 kb、 Xho Ⅰ 4 9 kb and Bam HⅠ 3 0 kb fragments by southern blotting with Ac MNPV derived probe. After cloning of the three fragments, restriction enzyme analysis was used to construct their restriction map. Partly sequencing of the three fragments shows that the p 74 gene of Sl MNPV has 69% of nuceotide identity and 66% of amino acid identity with that of Ac MNPV .
