摘要 随着基因在分子水平上的研究越来越深,基因检测也越来越多地被临床广泛应用,基因检测技术也从第一代测序技术逐步发展为第二代、第三代和第四代测序技术。第一代测序技术结果稳定,是基因诊断的金标准,弊端是高成本,低通量;第二代测序技术在第一代测序基础上提高了通量,是临床上应用最广的一种测序方法;第三代测序技术通量更加高、读长更加长、时间更短、成本更低;第四代测序技术还在实验阶段。本文分析以上四种基因的测序技术,浅谈其优点和弊端以及在医学检验中的应用。 Along with the deeper and deeper research of gene on the molecular level,gene detection is more and more widely used in clinic. Gene detection technology has gradually developed from the first to the fourth generation sequencing technology. The first generation of sequencing technology is stable,and used as the gold standard of gene diagnosis,the disadvantage is high cost and low flux. The second generation sequencing technology has increased on the basis of the first generation sequencing,and is one of the most widely used sequencing methodsin clinic. The third generation sequencing technology has higher throughput,with longer reading length,shorter time and lower cost. The fourth generation of sequencing technology is still in the experimental stage. In this paper,four gene sequencing techniques were analyzed,and their advantages and disadvantages as well as their applications in medical testing were discussed.
