摘要
目的评价不同孔径三维立体泡沫状外消旋聚乳酸材料(PDLLA)的成骨能力。方法采 用溶剂注模并盐结晶颗粒沥滤法制成3种(70~150μm,150~300μm和400~700μm)孔径 的外PULLA料,在62例1cm兔桡骨去骨膜缺损模型中分别植入3种孔径材料和空白对照,术后2 、4、8、12周行X线、组织学观察及8、12周扫描电镜观察骨缺损区骨生成情况。结果70~15 0μm 与150~300μm材料成骨较好,与400~700μm组和对照组相比差异有显著性(P< 0.05 )。结论PDLLA具有良好的生物相容性和可吸收性,制成泡沫状具有较好的骨传导性能。
Objective To evaluate the effect of three-dimensional porous poly-DL -lactide(PDLLA)with different pore sizes on bone formation ,and to sift a val uble carrier for bone growth factors and scaffold for cell culture.Method PDLLA with three different pore sizes(70~150μm, 150~300μm and 400~700μm) were fa bricated by a solvent-casting particulate-leaching technique with NaCl as the leachable component. The materials above were implants into 1.0cm radial defects of 62 rabbits. The specimens were harvested in 2,4,8,12 weeks. The samples were examined by X-ray, histological and scanning electronic microscope(SEM).Result The materials having pore sizes in the range of 70~150 or 150~300μm stimulat ed more bone formation than the material with pore size in the range of 400~700 μm and control group. The former two experiment groups were significantly highe r compared with the latter two groups in every stage of postimplantation.Conclus ion PDLLA has good biocompatibility and absorbability. It has good osteoconducti bility when fablicated into porous material. Pore size and porosity significantl y affected the ability of osteoconduction. The foams with pore size in range of 70~300μm have better osteoconductibility.
