摘要
介绍了用液相色谱法测定蜂蜜中咖啡因含量 ,样品在碱性条件下经氯仿萃取 ,萃取液用HPLC进行检测 ,其线性范围在0.010×10 -6~10×10 -6(w) ,定量下限为0.010×10 -6(w) ,回收率>80% ,相对标准偏差<2%。
A method for the determination of caffeine in honey by high performance liquid chromatography was developed. Caffeine in honey sample was extracted by chloroform in alkaline medium. Chromatographic separation was performed in SUPELCO Discovery C16 column(25 cm×4.6 mm ID, 5 μm) with eluate of acetonitrile-water-acetic acid(16∶83∶1 by volume) system(flow rate 0.5 mL/min). The caffeine was detected with UV detector at 273 nm. The linear range of 0.010×10-6~10×10-6(w), lower limit of detectability of 0.010×10-6 (w), recovery above 80% and RSD below 2%(n=6) were obtained for the method.
