摘要
该实验从猪小肠中提取α-淀粉酶,对α-淀粉酶进行硫酸铵盐析.考察了最佳反应条件为41℃,pH6.6.并用海藻酸钙固定化酶,测定阿卡波糖对固定化α-淀粉酶的IC50为15.625μg/mL.在96孔板微反应体系中建立了α-淀粉酶抑制剂的固定化酶筛选模型,对甘草、绿茶、枸杞、龙眼核、桑叶和桑白皮等六种中草药的水提部分和醇提部分进行筛选,结果表明:甘草,桑白皮,桑叶的水提和醇提及龙眼的水提物对α-淀粉酶的抑制效果较明显.
α-Amylase is extracted from pig's small intestine and is partially purified with(NH4)2SO4 precipitation.The optimal reaction condition of α-amylase is obtained at 41℃ and pH 6.6.The enzyme is then immobilized with calcium alginate and the IC50 of acarbose to immobilized α-amylase is found to be 15.625 μg/mL.The screening model of α-amylase inhibitors is established through 96-well plates with immobilized enzymes.Using this model,the aqueous extract and ethanol extract of Liquorice,green tea,Chinese Wolfberry,Longan nut,Mulberry Leaf,White Mulberry Root-bark are screened.The results show that the aqueous extract and ethanol extract of Liquorice,Mulberry Leaf,and White Mulberry Root-bark have strongly α-amylase inhibition.
