摘要
目的 :探讨丹参抗肝纤维化的可能机制。方法 :链蛋白酶、胶原酶离体灌注大鼠肝脏 ,Nycodenz密度梯度离心分离肝星状细胞 (HSC)体外培养。不同浓度的丹参处理 ,用RT -PCR法半定量检测HSC的转化生长因子 β1(TGF - β1)mRNA与Ⅰ型胶原mRNA的表达。结果 :丹参对HSC的TGF - β1与Ⅰ型胶原mRNA的表达有抑制作用。结论 :丹参抗肝纤维化作用可能与TGF - β1和Ⅰ型胶原mRNA的表达下调有关。
Objective:To investigate the possible mechanism of antifibrotic action of Salvia miltiorrhiza at molecular level.Methods:Rat hepatic stellate cell was separated from rat liver by pronase E and collagenase perfusion and density gradient centrifugation with Nycodenz. Various concentrations of Salvia miltiorrhiza were added and incubated. The mRNA expressions of transforming growth factorβ 1 (TGF-β 1) and type Ⅰ procollagen were analyzed by RT-PCR.Results:Salvia miltiorrhiza could inhibit the expressions of TGF-β 1 and type Ⅰ procollagen mRNA.Conclusion:The antifibrotic action of Salvia miltiorrhiza may be related to the down-regulation of TGF-β 1 and type Ⅰ procollagen mRNA.
