家兔实验性心肌缺血肌动蛋白免疫组化定量研究

摘要 本文通过建立家兔心肌急性缺血的动物模型 ,应用免疫组织化学方法和图像分析技术 ,结合HE和PAS染色对心肌细胞肌动蛋白在心肌缺血时的早期改变进行了观察和测定。当心肌缺血 30min时 ,肌动蛋白发生了缺失 ,因此可以作为心肌早期缺血的指标。同时观察到在心肌缺血 1h内 ,肌动蛋白的阳性面积和强度随缺血时间的延长而显著降低 ,可较准确客观地判定心肌缺血时间。 The early histochemical changes of actin in ischemic myocardium following coronary artery occlusion in 18 rabbits were studied by histochemical, immunohistochemical and image analysis techniques. The resulrs shoued that the amount and the IRS of actin were significantly diminished 30min after ischemia (P<0.05). 60min after occlusion, the intensities and areas of actin were progressively markedly reduced. It is suggested the immunohistochemical changes of action may be used as an early criterion for acute myocardial ischemia, and it provides an accurate method for detemining the stage of early myocardial ischemia.
