摘要
为了研究山羊紧密连接相关蛋白ZO-1基因编码蛋白的组成结构及生物学特性,试验应用生物信息学方法对ZO-1蛋白的氨基酸序列组成、理化性质、亲疏水性、信号肽、跨膜结构域、磷酸化位点、亚细胞定位、蛋白质二级和三级结构进行预测与分析,并构建系统进化树。结果表明:该基因全长1 164 bp,编码387个氨基酸; ZO-1蛋白分子质量为45. 25 ku,理论等电点为6. 52,由20种氨基酸组成,呈酸性,不稳定指数为58. 54,是一个不稳定的亲水性蛋白;具有41个潜在的磷酸化位点,不存在信号肽和跨膜区;亚细胞定位分析显示,ZO-1蛋白主要存在于细胞核,在线粒体和细胞质中也有少量分布;二级结构预测显示,ZO-1蛋白以α-螺旋为主要结构,比例为68. 73%;三级结构预测显示,ZO-1蛋白是由α-螺旋、β-转角、β-折叠和无规卷曲组合成的超二级结构,经折叠、弯曲等一系列复杂过程形成了一个稳定的结构;系统进化树分析发现,山羊的ZO-1基因编码蛋白序列首先与绵羊聚为一类,然后与牛聚为一类,这与动物学分类结果一致,这种同源性在一定程度上代表着物种亲缘关系的远近。说明ZO-1基因有望成为维持山羊瘤胃上皮组织屏障功能和预防亚急性瘤胃酸中毒疾病的重要基因。
The aim of the present study was to explore the composition structure and biological characteristics of ZO-1 protein related to tight junction in goat. The amino acid sequence composition, physicochemical properties, hydrophilicity and hydrophobicity, signal peptide, trans- membrane domain, phosphorylation site, subcellular localization, secondary structure and tertiary structure of ZO- 1 protein were predicted and analyzed by bioinformatics method. And the phylogenetic tree was constructed. The results showed that the gene was 1 164 bp in full length and encoded 387 amino acids. The molecular weight of the encoded protein was 45.25 ku. The theoretical isoelectric point was 6.52. It was com- posed of 20 amino acids and was acidic. The instability index was 58.54. It was an unstable hydrophilic protein. The protein has 41 phosphoryl- ation sites and there was no signal peptide or transmembrane domains. Subcellular location analysis showed that the protein was mainly located in the cell nucleus and also distributed in mitochondria and cytoplasm. Secondary structure was mainly Alpha-helix (68.73%). The prediction of tertiary structure showed that ZO-1 protein was a super secondary structure composed of or-helix, [3-rotation angle, [3-sheet and random coil, which formed a stable structure through a series of complex processes, such as folding and bending. Phylogenetic tree analysis showed that goat ZO-1 gene coding protein sequence was first clustered with Ovis aries, and then clustered with cattle, which was consistent with zoological tax- onomy. To some extent, the homology of ZO- I gene coding protein sequence of goat and cattle represented the distant and near relationship of the species. These results suggest that ZO- 1 gene may be an important gene in maintaining rumen epithelial barrier function and preventing sub- acute tureen acidosis in goats.
