摘要
目的 探讨细胞周期相关蛋白syk、P57、ki- 67及cyclin D1在胃肠道间质瘤(GIST)中的表达及意义。 方法 随机挑选包头医学院第二附属医院和包头市肿瘤医院2013年1月至2015年12月正常人胃和肠黏膜各20例为对照组;胃和肠 GIST 标本80例为实验组,其中包括低级别胃和肠GIST各30例、高级别胃和肠GIST各10例。通过免疫组化s- p法对所选标本进行CD117及CD34检测,确保所选标本的准确性。利用免疫组化法检测20例正常胃和肠黏膜,80例GIST中syk、P57、ki- 67及cyclin D1的表达。 结果 所选标本除对照组外均同时表达CD117及CD34。对照组中 ki- 67、P57、cyclin D1表达较低或不表达,而syk呈高表达。实验组低度风险性GIST与对照组相比ki- 67、syk及p57表达在两组间无明显差异( P 〉0.05),而cyclin D1较对照组表达明显升高( P 〈0.05)。与对照组相比,实验组高级别GIST ki- 67、p57及cyclin D1表达明显升高,且均高于实验组低级别GIST中的表达( P 〈0.05)。而syk在高级别GIST表达减低或不表达( P 〈0.05)。 结论 与对照组正常胃肠黏膜相比,实验组GIST细胞周期相关蛋白ki- 67、P57、cyclin D1表达升高,且随着级别的升高而升高,而 syk表达则相反。由此可见,上述细胞周期蛋白在 GIST发病中扮演着重要角色,因此,探讨细胞周期相关分子机制,对理解GIST发病机制有着十分重要的意义。
Objective The paper aims to investigate the expressions and significance of cell cycle- related protein syk,P57,ki- 67 and cyclin D1,in gastrointestinal stromal tumors (GISTs). Methods Randomly selected 20 samples of normal gastric and intestinal mucosa respectively as the control group since 2013,and 80 samples of gastrointestinal stromal tumor specimens as experimental group,including 30 samples of the low- grade gastric stromal tumors and intestinal stromal tumors respectively,as well as 10 samples of the high- grade gastric stromal tumors and intestinal stromal tumors,respectively. CD117 and CD34 were detected by immunohistochemical S- P method to ensure the accuracy of the selected samples. The expression of P57, Syk,P57,Ki- 67 and cyclinD1 in 20 samples of normal gastric and intestinal mucosa and 80 samples of gastrointestinal stromal tumors were detected by immunohistochemistry method. Results CD117 and CD34 were simultaneously expressed in selected samples,except in the control group.In control group,the expressions of Ki- 67,cyclin D1 are low, P 〈0.05,while P57 and syk showed high expression, P 〈0.05.For the samples in experimental group:compared with the samples in the control group,in low- risk gastrointestinal stromal tumors,there was no significant difference in the expression of Ki- 67,syk and p57,P 〉 0.05, while cyclin D1 expression increased P 〈 0.05. Compared with samples in control group, the expression of both Ki-67 and eyelin D1 in high-risk gastrointestinal stromal tumors was significantly higher, than in low-risk gastrointestinal stromal tumors, P 〈 0.05. The expressions of p57 and Syk in high-risk gastrointestinal stromal tumors decreased or were not expressed,P 〈 0.05. Conclusions Compared with the samples in control group,the expression of cell cycle-related protein Ki-67 and eyelin D1 in gastrointestinal stromal tumors increased, along with the rise of risk level. The expression of P57 and syk decreased or not expressed in high-risk gastrointestinal stromal tumors. S
