法尼基转移酶在大鼠压力超负荷诱导的射血分数保留型心力衰竭心室重构中的作用

摘要 目的探讨法尼基转移酶对压力超负荷诱导的射血分数保留型心力衰竭(HFpEF)的作用。方法将30只SD大鼠随机分为手术组、法尼基转移酶抑制剂组(FTI组)和假手术组,每组10只,干预8周,颈动脉插管测血压,超声心动图评价左心室肥厚及舒张功能,左心室组织病理染色检测其重塑情况,G-LISA试剂盒检测左心室组织Ras蛋白活性。结果与假手术组比较,手术组血压明显升高,左心室舒张末期厚度明显升高,心肌细胞横截面积明显增大,心肌内血管周围纤维化明显增强,左心室Ras蛋白活性增强,差异有统计学意义(P<0.05)。与手术组比较,FTI组左心室舒张末期厚度、心肌细胞横截面积、心肌内血管周围纤维化均有所改善,左心室Ras蛋白活性下降,差异均有统计学意义(均P<0.05)。结论法尼基转移酶与大鼠压力超负荷诱导的HFpEF心肌重塑有关。 Objective To explore the effects of farnesyltransferase (FTase) on cardiac remodeling of rat with pressureoverload-induced heart failure with preserved ejection fraction (HFpEF). Methods 30 male SD rats were randomlydivided into abdominal aortic coarctation (ACC) group, FTase inhibition (FTI) group, Sham group, with 10 rats in eachgroup. Blood pressure was monitored with carotid catheterization and cardiac function was evaluated byechocardiography at 8 weeks post-operatively. The remodeling of left ventricle was evaluated by histology stain andcardiac Ras activity was measured by G-LISA assay kit. Results Blood pressure, left ventricular end-diastolic wallthickness, myocardial cross section, peri-vessel fibrotic tissue and Ras activity increased significantly in ACC groupcompared with sham group (all P〈0.05). The later four parameters increased less in FTI group than in ACC group (all P〈0.05). Conclusion FTase is related to cardiac remodeling of rat with pressure overload-induced HFpEF.
