纯钛表面加载氧化石墨烯-/银抗菌及细胞活力研究 被引量:1

Study on antibacterial activity and cell viability of pure titanium loaded with graphene oxide-/silver

摘要 目的:研究氧化石墨烯和银改性纯钛表面后对金黄色葡萄球菌形态及致病基因的影响。方法:通过电镀法和紫外还原法分别将氧化石墨烯和银加载于纯钛金属表面,利用场发射扫描电子显微镜、显微拉曼光谱仪、X射线光电子能谱仪和接触角测试仪对改性前后钛片表面形貌、性状及亲水性能进行表征;以金黄色葡萄球菌为目标菌,与改性前后3种形貌纯钛共培养,通过场发射扫描电子显微镜和实时定量荧光PCR对其形态及致病基因表达进行检测;利用CCK-8试剂盒检测各组试样表面前体成骨细胞活力水平。结果:通过电镀法和紫外还原法可成功将氧化石墨烯和银加载于纯钛金属表面,并提高其表面粗糙度和亲水性能,且不同形貌钛片对金黄色葡萄球菌形态和致病基因表达造成影响。另外,氧化石墨烯具有促进细胞活力水平作用,而氧化石墨烯和银降低细胞活力,但与纯钛片组相比,差异无统计学意义。结论:氧化石墨烯改性纯钛片后具有促进细菌黏附和细胞活力水平作用,而氧化石墨烯-银则具有显著抗菌效果。 Objective: To evaluate the effects of graphene oxide and silver modified titanium surface on the morphology and pathogenic genes of Staphylococcus aureus. Methods: Graphene oxide and silver were loaded on the pure titanium surface via electroplating method and ultraviolet reduction method,respectively,and the morphology,properties and hydrophilic properties were measured by field emission scanning electron microscope,micro raman spectrometer,X-ray photoelectron spectrometer,and contact angle tester,respectively. In addition,Staphylococcus aureus were utilized to be as the target bacteria,and co-cultured with three kinds of pure titanium surface. Then the morphology and pathogenic gene expression were detected by field emission scanning electron microscopy and real-time quantitative PCR. And CCK-8 kit was utilized to detect the viability of pre-osteoblasts on the specimen surface. Results: Graphene oxide and silver were successfully loaded onto pure titanium surface via electroplating and ultraviolet reduction method. The roughness and hydrophilic properties of specimens were improved. Afterwards,the morphology and pathogenic genes of Staphylococcus aureus on different titanium surface were affected. In addition,graphene oxide had the effect of promoting cell viability. However,graphene oxide and silver reduced cell viability. But there was no statistical difference. Conclusion: Pure titanium substrate modified by graphene oxide can promote bacterial adhesion cell viability. Moreover,Graphene oxide-silver has obvious antibacterial property.
